体外血管形成实验能够很好的模拟肿瘤血管发生过程,并且适合研究某些基因或药物对这一过程的影响。
吉凯基因提供的血管形成实验能通过血管内皮细胞(HUVEC)在基质胶(Matrigel)上形成血管的能力来判断某些基因或药物对血管形成的影响。
用待研基因shRNA慢病毒感染Panc-1细胞,3天后收集细胞上清,用目的细胞上清重悬细胞血管内皮细胞,将细胞悬液铺于基质胶上,4小时后观察血管形成能力的变化,实验结果表明干扰目的基因后,Panc-1细胞血管形成能力显著下降表明待研基因与Panc-1细胞血管形成显著相关。
